Neuroscience

Neuroplasticity & Ceremony

The brain can change. This page lays out what was measured after psilocybin, in whom, and for how long, and where the evidence stops.

What is neuroplasticity?

Neuroplasticity is the brain's ability to reorganize itself by forming new neural connections throughout life. It is how we learn, and how habits and trauma responses can change over time.

Plasticity is generally higher in childhood, which is one reason changing deeply ingrained patterns in adulthood can be hard. Researchers are studying whether psychedelics can temporarily reopen some of that flexibility. The evidence so far is preclinical or comes from small human studies.

What was measured, in whom, for how long

  • Mice, within 24 hours. A single psilocybin dose raised dendritic spine density and size by about 10% in frontal cortex, and the change persisted at one month (Shao et al., Neuron 2021). This is a preclinical finding.
  • Mice, weeks. A social-reward-learning critical period stayed open at two weeks after psilocybin and had closed by three weeks (Nardou et al., Nature 2023). Also preclinical.
  • Cultured and rodent neurons. Psychedelics increased neurite growth and spine formation through 5-HT2A, TrkB and mTOR signaling (Ly et al., Cell Reports 2018).
  • Patients with depression, about 3 weeks. fMRI measured increased global brain integration about three weeks after psilocybin therapy (Daws et al., Nature Medicine 2022).
  • Healthy volunteers, up to 1 month. Emotion and brain-function changes were reported up to one month after a single high dose (Barrett et al., Scientific Reports 2020), and patients with depression showed cognitive-flexibility gains at four weeks (Doss et al., Translational Psychiatry 2021).

Sources: Shao et al., “Psilocybin induces rapid and persistent growth of dendritic spines in frontal cortex in vivo,” Neuron 109(16):2535-2544, 2021 (doi:10.1016/j.neuron.2021.06.008); Nardou et al., Nature 618:790-798, 2023 (doi:10.1038/s41586-023-06204-3); Ly et al., Cell Reports 23(11):3170-3182, 2018 (doi:10.1016/j.celrep.2018.05.022); Daws et al., Nature Medicine28:844-851, 2022 (doi:10.1038/s41591-022-01744-z).

These are heterogeneous, indirect markers from preclinical or small studies. No study has measured a plasticity “window” of a particular length in humans, human structural plasticity has not been measured directly, and a BDNF increase after psilocybin has not been established in humans. Whether spine growth depends on 5-HT2A signaling or on direct TrkB binding is an open question (Vargas et al., Science 2023; Moliner et al., Nature Neuroscience 2023).

Where integration fits

No trial has tested whether integration support lengthens psilocybin's benefits. Ceremonia's integration curriculum, IFS parts work, somatic titration, journaling and lifestyle practices, begins in the days immediately following ceremony. That timing is a matter of our own practice, not a research finding.

The research tells us what changed in the studied groups. It does not promise any outcome at a retreat.

Neuroplasticity without psychedelics

Meditation, exercise, sleep quality, and social connection are associated with brain health, and cross-sectional studies of experienced meditators report modest and mixed structural differences. The Ceremonia preparation curriculum builds these practices before ceremony as part of our own approach.

Learning requires more than a window

Neuroplasticity describes the nervous system’s capacity to change. Researchers have observed changes in cellular and molecular markers after psychedelic exposure, much of this work in preclinical models. Those observations are interesting, but they do not establish that a person will acquire a particular habit or that every change is beneficial.

Imagine deciding to speak more openly with a friend. A meaningful experience might make that possibility feel salient, but the conversation still depends on timing, consent, practice, and the friend’s response. Integration can offer a place to reflect and choose a small action; it is not a guaranteed method for rewiring the brain.
